General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. Syntaxin-1A (UniProt: Q16623; also known as Neuron-specific antigen HPC-1) is encoded by the STX1A (also known as STX1) gene (Gene ID: 6804) in human. This gene encodes a member of the syntaxin superfamily. Syntaxins are nervous system-specific proteins implicated in the docking of synaptic vesicles with the presynaptic plasma membrane. Synaptic vesicles store neurotransmitters that are released during calcium-regulated exocytosis. The specificity of neurotransmitter release requires the localization of both synaptic vesicles and calcium channels to the presynaptic active zone. Syntaxins also binds synaptotagmin in a calcium-dependent fashion and interact with voltage dependent calcium and potassium channels via the C-terminal H3 domain. This gene product is a key molecule in ion channel regulation and synaptic exocytosis. Alternatively spliced transcript variants encoding three different isoforms have been found for this gene. Syntaxin-1A is highly expressed in embryonic spinal cord and ganglia and, in adult cerebellum and cerebral cortex. Isoform 2 is found to be expressed in heart, liver, fat, skeletal muscle, kidney and brain. Structurally, STX1A is a single-pass type IV membrane protein and consists of a coiled coil domain (aa 68-109), t-SNARE coiled-coil homology domain (aa 192-254) and, possess a single C-terminal transmembrane domain facilitating its anchorage to the presynaptic membrane and interaction with other SNARE proteins like SNAP-25 and VAMP-2. Dysregulation of STX1A has been implicated in various neurological disorders, including epilepsy and schizophrenia, highlighting its role in synaptic plasticity and neurotransmission. Recent Study reported that deletion of syntaxin-1A and B in mice can cause embryonic lethality and, leads to gross morphological abnormalities and neuronal cell loss. Furthermore, STX1A mutations and aberrant localization have been linked to the pathogenesis of neurodegenerative diseases, underscoring its importance in maintaining synaptic integrity and function. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human Syntaxin-1A.

Application

Quality Control Testing

Evaluated by Western Blotting in Neuro-2a cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Syntaxin-1A in Neuro-2a cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Syntaxin-1A in lysates from PC-12 cells, human brain tissue, and mouse brain tissue.

Affinity Binding Assay: A representative lot of this antibody bound Syntaxin-1A peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Syntaxin-1A in human cerebral cortex tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2B21 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects Syntaxin-1A. It targets an epitope within 22 amino acids from the C-terminal half. This antibody may cross react to Syntaxin-1B.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
