General description
Serine incorporator 5 (UniProt:Q86VE9) is encoded by the SERINC5 (also known as C5orf12 ) gene (Gene ID: 256987) in human. SERINC5 is a member of the serine incorporator family, is characterized by its role as a multipass transmembrane protein involved in lipid metabolism and antiviral defense. Its a non-ATP-dependent, non-specific lipid transporter for phosphatidylserine, phosphatidylcholine, and phosphatidylethanolamine. It also functions as a scramblase that flips lipids in both directions across the membrane. Structurally, SERINC5 contains approximately 10 to 11 transmembrane domains, enabling its localization predominantly at the plasma membrane, where it plays a critical role in inhibiting viral entry by disrupting the fusion of retroviral particles with host cells. SERINC5 is highly expressed in various tissues including placenta, skeletal muscle, spleen, thymus, testis and peripheral leukocyte and, is known to be weakly expressed in the heart, liver and fetal brain. Four isoforms of SERINC5 have been described that are produced by alternative splicing. Its involvement in disease is particularly significant in the context of HIV-1 infection, where the viral protein Nef counteracts SERINC5′s antiviral effects by redirecting it to lysosomes for degradation, thereby enhancing viral infectivity. Furthermore, mutations and dysregulation of SERINC5 have been associated with several pathologies, including neurodegenerative diseases and certain cancers. Evaluated by Western Blotting in A-431 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SERINC5 in A-431 cell lysate.
Tested Applications
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected SERINC5 in A-431 cells.
Affinity Binding Assay: A representative lot of this antibody bound SERINC5 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected SERINC5 in human prostate tissue sections.
