General description
Phosphoglycerate kinase 1 (UniProt: P00558; also known as Cell migration-inducing gene 10 protein, Primer recognition protein 2, PRP 2, EC:2.7.11.1, EC:2.7.2.3) is encoded by the PGK1 (also known as PGKA) gene (Gene ID: 5230) in human. A pseudogene of PGK1 gene has been found on the X-chromosome and another on chromosome 19 and 21. Structurally, PGK1 is characterized by a highly conserved domain architecture, facilitating its function as both a glycolytic enzyme that catalyzes the conversion of 1,3-diphosphoglycerate to 3-phosphoglycerate and, a protein kinase when localized to mitochondria, where it phosphorylates pyruvate dehydrogenase kinase 1, thereby regulating cellular metabolism and promoting glycolysis over oxidative phosphorylation. The encoded protein has been identified as a moonlighting protein based on its ability to perform mechanistically distinct functions. The protein may also act as a cofactor for polymerase alpha. PGK1 exhibits a ubiquitous expression profile across various tissues. Additionally, this protein is secreted by tumor cells where it participates in angiogenesis by functioning to reduce disulfide bonds in the serine protease, plasmin, which consequently leads to the release of the tumor blood vessel inhibitor angiostatin. Two isoforms of PGK1 have been described that are produced by alternative splicing. Deficiency of the enzyme is associated with a wide range of clinical phenotypes hemolytic anemia and neurological impairment. Dysregulation of PGK1 is implicated in several diseases, including phosphoglycerate kinase deficiency, which manifests as hemolytic anemia and neurological impairments due to mutations affecting PGK1 expression and function. Studies suggested that activation of Pgk1 in cytosol may lead to a reduction in protein aggregation across various neurodegenerative conditions. Western Blotting Analysis: A 1:10,000 dilution of this antibody detected PGK1 in U-937 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PGK1 in lysate from HT-29 cells.
Affinity Binding Assay: A representative lot of this antibody bound PGK1 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PGK1 in HeLa cells.
