General description

Guanine nucleotide-binding protein G(I)/G(S)/G(T) subunit beta-1 (UniProt: P62873; also known as Transducin beta chain 1) is encoded by the GNB1 gene (Gene ID: 2782) in human. GNB1 is a critical component of the heterotrimeric G proteins, which are pivotal in the transduction of chemical signals from the outside to the inside of cells. It forms a complex with G protein gamma subunits (G ) and interacts with G protein alpha subunits (G ) to transmit signals from G protein-coupled receptors (GPCRs) to intracellular effector pathways. GNB1 consists of seven WD repeat domains that are responsible for the structural integrity and protein-protein interactions necessary for signal transduction. GNB1, as part of the Gβ subunit in heterotrimeric G proteins, plays a central role in mediating signal transduction from a wide variety of GPCRs to downstream signaling pathways. This subunit helps in the regulation of several key cellular processes, including cell growth, hormone secretion, and neural transmission. It affects various signaling pathways by switching between active and inactive forms of G subunits, thus modulating the activity of downstream effectors such as adenylate cyclase and phospholipases. Mutations and dysregulation of GNB1 have been linked to a variety of diseases. Notably, GNB1 mutations can lead to GNB1 syndrome, characterized by developmental delays, neurological issues, and distinct facial features. These mutations disrupt normal G protein signaling, leading to altered cellular responses and developmental abnormalities. Additionally, aberrant GNB1 signaling is implicated in cancer, where it can affect cell proliferation and survival. It also plays a role in cardiovascular diseases through its impact on heart rate and blood pressure regulation. KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal region of human GNB1.

Application

Quality Control Testing

Evaluated by Western Blotting in C6 cell lysate.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected GNB1 protein in C6 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected GNB1 in lysates from mouse brain and human brain tissues and, a 1:1000 dilution from a representative lot detected GNB1 in Jurkat cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound GNB1 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected GNB1 in human cerebral cortex tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GNB1 in C6 and SH-SY5Y cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1J8 is a ZooMAb® recombinant rabbit monoclonal antibody that detects GNB1. It targets an epitope within 16 amino acids from the N-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
