General description
Integrin alpha-E (UniProt: P38570; also known as HML-1 antigen, Integrin alpha-IEL, Mucosal lymphocyte 1 antigen, CD103) is encoded by the ITGAE gene (Gene ID: 3682) in human. Integrin alpha-E (CD103) is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 19-1124), a transmembrane domain (aa 1125-1147), and a short cytoplasmic domain (aa 1148-1179). It is expressed on a subclass of T-lymphocytes known as intra-epithelial lymphocytes that are located between mucosal epithelial cells. CD103 expression has been reported on CD4+ regulatory T cells and about 4% of circulating CD8+ T cells also bear the CD103 molecule. Expression of CD103 on alloreactive cells is shown to be augmented by IL-4, IL-10, or TGF-β and decreased by addition of IL-12. It forms a heterodimer with β-7 subunit to form a receptor for E-cadherin and mediate adhesion of intra-epithelial T cells to epithelial cell monolayers. Clone Ber-ACT8 is shown to react with in vitro activated T cells and a small subset of normal resting T cells, but not with resting B cells or several other human cell lines. Its reactivity is restricted to a few T cells in the peripheral blood, the extrafollicular areas of lymph nodes and tonsils, and splenic red pulp. Application
Quality Control Testing
Evaluated by Western Blotting in Jurkat cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD103 in Jurkat cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected His-tagged, recombinant human CD103 (Integrin E) and β7.
Flow Cytometry Analysis: 1 µg from a representative lot detected CD103 in one million Jurkat cells.
Affinity Binding Assay: A representative lot of this antibody bound CD103 peptide with a KD of 3.2 x 10-7 in an affinity binding assay.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1I14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects CD103. It targets an epitope within 25 amino acids from the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
