Specificity

Clone 1F22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects GAL4. It targets an epitope within 22 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal region of human Regulatory protein GAL4 (GAL4).

Application

Quality Control Testing

Evaluated by Western Blotting with GST-tagged recombinant fragment of yeast GAL4.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected GST-tagged recombinant fragment corresponding 114 amino acids from the C-terminal region of yeast GAL4 expressed in E.coli.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected yeast GAL4 spiked with GST-tagged recombinant fragment of yeast GAL4.

Affinity Binding Assay:: A representative lot of this antibody bound GAL4 peptide with a KD of 8.3 x 10-7 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: Serial of dilutions from a representative lot detected GST-tagged recombinant fragment corresponding to 114 amino acids of yeast GAL4.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Regulatory protein GAL4 (UniProt: P04386; also known as GAL4) is encoded by the GAL4 (also known as YPL248C) gene (Gene ID: 855828) in Saccharomyces cerevisiae (yeast). GAL4 is a zinc-binding nuclear protein that binds to DNA as homodimer and its DNA binding region is localized within the first 74 amino acids. It acts as a positive regulator of gene expression of the galactose induced genes such as GAL1,GAL2, GAL7, GAL10, and MEL1, which code for enzymes used to convert galactose to glucose. It recognizes a 17 base pair sequence (5′-CGGRNNRCYNYNCNCCG-3′) the upstream activating sequence (UAS-G) of these genes. GAL4 is reported to bind to four sites in the UAS-G sequence. It contains a nine amino acids TAD motif (aa 862-870) that serves as a transactivation domain, which is present in a large number of yeast and animal transcription factors. The yeast GAL4 is shown to activate transcription in Drosophila but only from promoters that bear GAL4 binding sites. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonal. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
