General description
Actin-related protein 3 (UniProt: P61158; also known as Actin-like protein 3, ) is encoded by the ACTR3 (also known as ARP3) gene (Gene ID: 10096) in human. Actin-related protein 3 (ARP3) is a critical component of the Arp2/3 complex, a highly conserved multiprotein assembly involved in actin cytoskeleton dynamics. ARP3 shares significant homology with actin and functions as part of the complex to nucleate the formation of branched actin filaments. This nucleating activity occurs at a 70° angle to pre-existing filaments, enabling cellular protrusions such as lamellipodia and filopodia, essential for cell motility. ARP3 regulates diverse cellular processes, including actin polymerization, intracellular trafficking, phagocytosis, and endocytosis. It is critical for maintaining cellular polarity and is implicated in pathways such as actin nucleation via the WASP-Arp2/3 complex, tight junction formation, and terminal differentiation in epithelial cells. ARP3 also plays a role in DNA damage repair by promoting nuclear actin polymerization during homologous recombination. Expression of ARP3 is ubiquitous, with high levels observed in tissues actively remodeling their cytoskeleton, such as epithelial and immune cells. Mutations or dysregulation of the ACTR3 gene can lead to pathological conditions such as Wiskott-Aldrich Syndrome, characterized by defective actin polymerization in immune cells, and Lynch Syndrome 2, a cancer predisposition syndrome. Additionally, ARP3 expression influences tumor metastasis, particularly in hepatocellular carcinoma, where it is associated with poor prognosis. Application
Quality Control Testing
Evaluated by Western Blotting in Jurkat cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ARP3 in Jurkat cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ARP3 in rat spleen tissue lysate.
Immunocytochemistry Analysis: A 1:100 Dilution from a representative lot detected ARP3 in HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound with ARP3 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 1C7 is a ZooMAb® rabbit recombinant monoclonal antibody that detects ARP3. It targets an epitope within 21 amino acids from the internal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
