General description

Sorting nexin-27 (UniProt: Q96L92) is encoded by the SNX27 (also known as KIAA0488) gene (Gene ID: 81609) in human. This gene encodes a member of the sorting nexin family, a diverse group of cytoplasmic and membrane-associated proteins involved in endocytosis of plasma membrane receptors and protein trafficking through these compartments. All members of this protein family contain a phosphoinositide binding domain (PX domain). Structurally, SNX27 is approximately 62 kDa protein and has unique combination of a PDZ domain (aa 43-136) and a PX (phox) domain (aa 161-269). It is primarily localized to early endosomes and localizes to immunological synapse in T-cells. Expression studies indicate that SNX27 is widely expressed across various tissues, including the brain, kidney, liver, and in cells of hematopoietic origin. Three isoforms of SNX27 have been described that are produced by alternative splicing. SNX27 is involved in the modulation of signaling pathways, including the Ras-ERK pathway, which is crucial for T cell activation and function. SNX27′s interaction with diacylglycerol kinase has been shown to impact T cell receptor signaling and the immune synapse formation. Dysregulation of SNX27 has been implicated in several diseases, including various cancers however, the roles and underlying mechanisms governed by SNX27 in tumorigenesis remains to be poorly understood. Future studies are required to further evaluate and demonstrate the crucial role of SNX27 in cancer and to develop strategies to prevent and treat tumorigenesis. Specificity

Clone 1C16 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects SNX27. It targets an epitope within 23 amino acids from the N-terminal half. It is predicted to react with all isoforms.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the N-terminal half of human SNX27.

Application

Quality Control Testing

Evaluated by Western Blotting in NIH/3T3 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SNX27 in NIH/3T3 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected SNX27 in lysate from HepG2, C6, and A-431 cells.

Affinity Binding Assay: A representative lot of this antibody bound SNX27 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected SNX27 in human tonsil tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
