General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Histone deacetylase 11 (UniProt: Q96DB2; also known as EC:3.5.1.98; HD11) is encoded by the HDAC11 gene (Gene ID: 79885) in human. Histone deacetylase 11 (HDAC11) is the smallest and most recently identified member of the histone deacetylase family, classified as the sole representative of Class IV HDACs. Structurally, HDAC11 shares conserved features with other classical HDACs, including a zinc-dependent catalytic domain that is essential for its enzymatic activity. This domain forms a funnel-shaped active site responsible for deacetylating lysine residues on histones and other proteins, impacting chromatin remodeling and gene expression. HDAC11 localizes primarily to the nucleus but has also been detected in other subcellular compartments, suggesting functional versatility. Its expression is enriched in the brain, heart, and testis, with variable levels in immune cells and other tissues. HDAC11 is pivotal in regulating immune responses, acting as a negative regulator of interleukin-10 (IL-10) in antigen-presenting cells, thereby modulating inflammatory processes. It also influences the expression of pro-inflammatory cytokines, such as IL-6 and TNF- , and chemokines like CCL2, linking its activity to immune cell maturation, migration, and function. Beyond immunity, HDAC11 is implicated in cellular processes such as proliferation, differentiation, and lipid metabolism. Knockout studies indicate that HDAC11 deficiency enhances mitochondrial function, skeletal muscle regeneration, and remyelination in the central nervous system, highlighting its role in tissue repair and metabolic regulation. HDAC11 is part of a complex network, associating with other HDACs like HDAC6 and interacting with transcriptional machinery components. These interactions extend its influence beyond chromatin remodeling to processes like RNA splicing, protein ubiquitination, and signal transduction. Aberrant HDAC11 activity or expression has been implicated in various pathologies, including cancers and inflammatory disorders. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of human HDAC11.

Application

Quality Control Testing
Evaluated by Western Blotting in SK-BR-3 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected HDAC11 in SK-BR-3 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected HDAC11 in human brain tissue lysate.
Affinity Binding Assay: A representative lot of this antibody bound with HDAC11 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected HDAC11 in human testis tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1E11 is a ZooMAb® rabbit recombinant monoclonal antibody that detects HDAC11. It targets an epitope within 25 amino acids from the C-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
