General description

E-selectin (UniProt: P16581; also known as CD62E, CD62 antigen-like family member E, Endothelial leukocyte adhesion molecule 1 (ELAM-1), Leukocyte-endothelial cell adhesion molecule 2 (LECAM2)) is encoded by the SELE (also known as ELAM1) gene (Gene ID: 6401) in human. E-selectin is single-pass type I membrane glycoprotein, a member of the selectin family of adhesion molecules, characterized by an extracellular domain containing a C-type lectin domain (aa 22-139), an EGF-like domain (aa 140-175), and a short cytoplasmic tail. E-selectin plays a crucial role in the immune response by mediating the adhesion of leukocytes to endothelial cells during inflammation. Upon activation by pro-inflammatory cytokines (such as TNF- and IL-1), E-selectin is expressed on the surface of endothelial cells, allowing leukocytes to roll along the vessel wall and eventually migrate into tissues. It is not constitutively expressed but is upregulated during inflammatory processes, such as in response to cytokines. E-selectin expression can vary depending on tissue type and the presence of inflammatory signals. E-selectin is localized on the surface of endothelial cells. It is stored in Weibel-Palade bodies, which are specialized storage granules within endothelial cells, and is translocated to the cell surface upon activation by inflammatory mediators. E-selectin is implicated in various inflammatory and cardiovascular diseases. Elevated levels of E-selectin are associated with chronic inflammatory conditions, such as atherosclerosis, rheumatoid arthritis, and asthma. KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal half of human CD62E.

Application

Quality Control Testing

Evaluated by Western Blotting in human bone marrow tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD62E in lysate from human bone marrow tissue.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD62E in human bone marrow tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with CD62E peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1I3-H1 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CD62E. It targets an epitope within 17 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
