General description
Nucleobindin-1 (UniProt: Q02818; also known as NUC) is encoded by the NUCB1 (also known as CALNUC) gene (Gene ID: 4924) in human. Nucleobindin-1 (NUCB1) is a multifunctional calcium-binding protein predominantly localized in the Golgi apparatus. NUCB1 contains EF-hand motifs that undergo conformational changes upon calcium binding, facilitating protein-protein and nucleic acid interactions. It is also characterized by a signal peptide and a leucine zipper domain, which are critical for its function and localization. NUCB1 acts as a major calcium-binding protein involved in calcium homeostasis and intracellular signaling. NUCB1 serves as a non-receptor guanine nucleotide exchange factor (GEF), activating G-protein alpha subunits, which are essential for various signaling pathways. It plays a role in regulating the unfolded protein response (UPR) during endoplasmic reticulum (ER) stress by modulating the cleavage of ATF6, a key transcription factor in UPR signaling. NUCB1 has also been implicated in maintaining Golgi structure and function, as well as in neuronal processes, where it acts as a marker for neuronal cells. The expression of NUCB1 is observed across diverse tissue types, with enriched profiles in neuronal and immune-related cells. Dysregulation of NUCB1 has been associated with several diseases, including prediabetes syndrome and kidney renal clear cell carcinoma, where it is considered a prognostic marker. Its role in calcium signaling and cellular stress responses links NUCB1 to pathological conditions such as cancer and metabolic disorders. Immunogen
Application
Evaluated by Western Blotting in PC-3 cell lysate.
Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected NUCB1 in PC-3 cell lysate.
Tested Applications
Immunocytochemistry Analysis: A 1:1,000 Dilution from a representative lot detected NUCB1 in HepG2 cells.
Affinity Binding Assay: A representative lot of this antibody bound with NUCB1 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis (IHC(P)): A 1:100 dilution from a representative lot detected NUCB1 in human cerebral cortex tissue sections.
