General description
Coiled-coil-helix-coiled-coil-helix domain-containing protein 10, mitochondrial (UniProt: Q8WYQ3; also known as Protein N27C7-4) is encoded by the CHCHD10 (also known as C22orf16) gene (Gene ID: 400916) in human. CHCHD10 belongs to a family of mitochondrial proteins located in the intermembrane space that is enriched at cristae junctions. CHCHD10 displays a ubiquitous distribution pattern, but its expression is highest in heart and liver and lowest in spleen. It is involved in the maintenance of mitochondrial organization, mitochondrial cristae structure, and mitochondrial DNA stability. CHCD10 has a non-structured N-terminal region, followed by a highly hydrophobic helix and a C-terminal CHCH domain characterized by a Cx(9)C motif and 2 additional cysteines. The four cysteine residues (102, 112, 122 and 132) of the CHCH domain are involved in two disulphide bonds. Overexpression of a CHCHD10 mutant allele in HeLa cells is reported to lead to fragmentation of the mitochondrial network and ultrastructural major abnormalities including loss, disorganization and dilatation of cristae. Mutations in CHCHD10 gene are reported to cause frontotemporal dementia and/or amyotrophic lateral sclerosis 2 (FTDALS2). Specificity
Clone 3J12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CHCHD10. It targets an epitope within 14 amino acids from the C-terminal.

KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal of human CHCHD10.
Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CHCHD10 in HeLa cell lysate.
Tested Applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CHCHD10 in HeLa cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CHCHD10 in human liver tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
