General description
Caspase-6 (UniProt: P55212; also known as EC:3.4.22.59; CASP-6, CSP-6, Apoptotic protease Mch-2) is encoded by the CASP6 (also known as MCH2) gene (Gene ID: 839) in human. Caspase-6 is a cysteine protease classified as an effector caspase, playing key roles in apoptotic and inflammatory programmed cell death pathways, including apoptosis, pyroptosis, necroptosis, and the combined process known as PANoptosis. CASP6 is expressed as a dimeric zymogen composed of a short pro-domain (p11) and a large subunit (p18), with activation requiring cleavage at specific sites. Its activity is tightly regulated by upstream initiator caspases, particularly caspase-8, and by interactions with cellular scaffolding proteins such as RIPK3 and ZBP1, which mediate inflammasome activation and innate immune responses. Caspase-6 is involved in the degradation of key cellular targets, such as lamin A, which contributes to nuclear disassembly during apoptosis. Its roles extend to innate immunity, where it cleaves immunosuppressive molecules like interleukin-10 and IRAK-M, thus promoting immune activation. Caspase-6 is also implicated in neurodegenerative diseases through its cleavage of mutant huntingtin (mHTT) protein in Huntington s disease and amyloid precursor protein (APP) in Alzheimer s disease, leading to toxic protein aggregates that exacerbate disease progression. Caspase-6 is predominantly localized to the cytosol, although its activity is spatially regulated within cellular compartments during apoptosis and inflammatory responses. Emerging evidence highlights Caspase-6′s ability to regulate cancer cell invasion by modulating matrix metalloproteinases (MMP-2 and MMP-9), suggesting a broader involvement in tumor progression and metastasis.
