General description
Protein Wnt-3a (UniProt: P56704) is encoded by the WNT3A gene (Gene ID: 89780) in human. The evolutionarily conserved Wnt signaling pathway is involved in cell proliferation, fate and polarity determination during embryonic development, and in cell migration. Abnormalities in Wnt signaling are reported to promote birth defects, degenerative diseases, and cancer. Wnt genes encode a large family of secreted, cysteine-rich proteins of approximately 350 to 400 amino acids (~40 kDa) that are important in development and maintenance of adult tissues. Nineteen different Wnt proteins have been identified in mammals thus far. They are either tethered to the plasma membrane or exit the cell via multiple routes, including direct release from the plasma membrane by solubilization and the formation of exosomes. Wnt-3a serves as a ligand for the frizzled family of transmembrane receptors. It induces the accumulation of β-catenin, thereby activating the canonical Wnt signaling pathway that results in the activation of transcription factors of the TCF/LEF family. Wnt-3a can promote or suppress tumor progression via the canonical Wnt signaling pathway depending on cancer type. Wnt-3a treatment is reported to provide neuroprotection and promote neurogenesis and improve neurocognitive function in traumatic brain injury murine models.
