General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. ZooMAb® antibodies are reliably available and ready to ship when you need them.

Specificity

Clone 1H8-L1 is a ZooMAb® rabbit recombinant monoclonal antibody that detects IL-22. It targets an epitope within 21 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal half of human IL-22.

Application

Quality Control Testing

Evaluated by Western Blotting in human thymus tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IL-22 in human thymus tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected recombinant fragment of human IL-22.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IL-22 in human tonsil and human colon tissue sections.

Flow Cytometry Analysis: 1 µg of this antibody from a representative lot detected IL-22 in one million human peripheral blood mononuclear cells (PBMC) treated with 500 ng/mL Phorbol 12-myristate 13-acetate (PMA), 500 ng/ml ionomycin, 1 g/ml of Brefeldin A (4- 6 hours).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interleukin-22 (UniProt: Q9GZX6; also known as Cytokine Zcyto18, IL-10-related T-cell-derived-inducible factor (IL-TIF)) is encoded by the IL22 (also known as ILTIF, ZCYTO18, UNQ3099/PRO10096) gene (Gene ID: 50616) in human. IL-22 is an -helical cytokine that contributes to the inflammatory response in vivo. It is a secretory protein that is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to produce the mature protein. It is immediately produced to initiate immune response against several tissue impairments. It is considered as a fundamental mediator in inflammation, mucous production, protective role against pathogens, wound healing, and tissue regeneration. It is produced by several populations of immune cells, including macrophages, β T cells classes Th1, Th22, and Th17 cells at the site of inflammation. It is shown to up-regulate a number of mucus associated genes, such as MUC1, MUC3, MUC10, and MUC13 in colonic and tracheal epithelial cells. Along with IL-17A, it also produces pulmonary mucus as a control against Gram negative pathogens. Its biological activity is initiated by binding to a cell-surface complex composed of IL-22R1 and IL-10R2 receptor chains and further regulated by interactions with a soluble binding protein (IL-22BP). IL-22 is shown to have a very high affinity for binding to IL-22BP, which acts as a natural antagonist of IL-22. IL-22BP binding occurs at specific sites on IL-22 that are also utilized by IL-22R1. (Ref.: Arshad, T., et al. (2020). Front. Immunol. 11; 2148). This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
