General description
Phosphatidylinositol 3-kinase catalytic subunit type 3 (UniProt: Q8NEB9; also known as EC:2.7.1.137, PI3-kinase type 3;PI3K type 3, PtdIns-3-kinase type 3, Phosphatidylinositol 3-kinase p100 subunit, Phosphoinositide-3-kinase class 3, hVps34 ) is encoded by the PIK3C3 (also known as VPS34) gene (Gene ID: 5289) in human. It is a member of the PI 3-kinase family that mediates the active diversion of proteins from the secretory pathway to vacuoles. PIK3C3 is involved in both receptor-mediated signal transduction and intracellular trafficking. It enables 1-phosphatidylinositol-3-kinase activity and is Involved in several processes, including early endosome to late endosome transport, macroautophagy, and phosphatidylinositol-3-phosphate biosynthetic process. PIK3C3 acts upstream of or within autophagy and protein lipidation. It mediates catalysis on membranes and suppress futile adenosine triphosphatase cycles. Subcellularly, it is located in autolysosome, late endosome, and midbody. The protein appears to alternate between a closed cytosolic form and an open form on the membrane. The protein is ubiquitously expressed, with a highest expression in skeletal muscle. Dysregulation of PIK3C3 has been implicated in several diseases including glioma and also, acts as a critical driver in Cowden Syndrome (CS) and Cowden-like syndrome (CSL), highlighting its significance in oncogenic processes. Recent studies have elucidated its role in modulating cellular responses to stress and nutrient availability, further establishing PIK3C3 as a pivotal player in maintaining cellular homeostasis and implicating it in various pathologies associated with aberrant autophagic activity. Immunogen
Application
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected PIK3C3 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PIK3C3 in lysates from C6 cells and human brain tissue and, a 1:10,000 dilution from a representative lot detected GST-tagged, full-length recombinant human PIK3C3.
Affinity Binding Assay: A representative lot of this antibody bound PIK3C3 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Enzyme Immunoassay (ELISA): A representative lot of this antibody detected PIK3C3 protein in ELISA application.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
