General description

Interleukin-2 receptor subunit beta (UniProt: P14784; also known as IL-2 receptor subunit beta, IL-2R subunit beta, IL-2RB, High affinity IL-2 receptor subunit beta, Interleukin-15 receptor subunit beta, p70-75 (p75), CD122) is encoded by the IL2RB (also known as IL15RB) gene (Gene ID: 3560) in human. IL-2R is a non-covalent dimer of an alpha and a beta subunit. It exists in 3 different forms: a high affinity dimer, an intermediate affinity monomer (beta subunit), and a low affinity monomer (alpha subunit). The high and intermediate affinity forms also associate with a gamma subunit. Both the intermediate and high affinity forms of the receptor are involved in receptor-mediated endocytosis and transduction of mitogenic signals from interleukin 2. IL2RB is a single-pass type I membrane protein. It is synthesized with a signal peptide (aa 1-26), which is cleaved off to produce the mature form that has an extracellular domain (aa 27-240), a transmembrane domain (aa 241-265), and a long cytoplasmic domain (aa 266-551). IL2RB contains a WSXWS motif (aa 220-224), which is essential for its proper folding and thereby efficient intracellular transport and cell-surface receptor binding. The protein also has a Box 1 motif (aa 278-286) that is required for JAK interaction and/or activation. This receptor is predominantly expressed in T cells, but it is also found in other immune cells, including B cells and natural killer cells. Dysregulation of IL2RB expression or function has been implicated in several diseases, including autoimmune disorders and cancer. Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal, extracellular domain of human IL2RB.

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IL2RB in Raji cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IL2RB in lysates from HEK-293T cells and rat spleen tissue.

Affinity Binding Assay: A representative lot of this antibody bound IL2RB peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected IL2RB in human spleen tissue and human bone marrow tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 4D2 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects IL2RB. It targets an epitope within 22 amino acids from the N-terminal, extracellular domain.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
