Analysis Note

C-type lectin domain family 4 member C (UniProt: Q8WTT0; also known as Blood dendritic cell antigen 2 (BDCA-2), C-type lectin superfamily member 7, Dendritic lectin, CD303) is encoded by the CLEC4C (also known as BDCA2, CLECSF11, CLECSF7, DLEC, HECL) gene (Gene ID: 170482) in human. C-type lectin domain family 4 member C (CLEC4C) is a type II transmembrane glycoprotein belonging to the C-type lectin superfamily. It contains a carbohydrate recognition domain that facilitates binding to asialo-galactosyl-oligosaccharides. CLEC4C is predominantly expressed on plasmacytoid dendritic cells (pDCs), making it a highly specific marker for these cells. Its distribution is restricted to humans and plays a critical role in antigen capture and immune regulation. CLEC4C acts as an inhibitory receptor on pDCs by suppressing the production of type I interferons (IFN- /β) upon ligation. This mechanism is important for maintaining immune homeostasis and preventing excessive inflammatory responses. CLEC4C is involved in pathways such as the innate immune system and CLEC7A (Dectin-1) signaling, which are central to host defense against infections and immune regulation. Dysregulation of CLEC4C has been associated with autoimmune diseases such as systemic lupus erythematosus (SLE) and psoriasis, where impaired CLEC4C signaling leads to excessive interferon production. Moreover, CLEC4C expression is altered in blastic plasmacytoid dendritic cell neoplasms (BPDCN), a rare and aggressive hematological malignancy, where it serves as a diagnostic marker. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Application

Quality Control Testing

Evaluated by Western Blotting in K-562 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CLEC4C in K-562 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CLEC4C in lysate from human lymph node tissue and human tonsil tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CLEC4C in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with CLEC4C peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

General description

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal extracellular domain of human CLEC4C.

Other Notes

Clone 2O23 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CLEC4C. It targets an epitope within 22 amino acids from the N-terminal extracellular domain.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
