General description

Alpha-crystallin B chain (UniProt: P02511; also known as Alpha(B)-crystallin, Heat shock protein beta-5 (HspB5), Heat shock protein family B member 5, Renal carcinoma antigen NY-REN-27, Rosenthal fiber component) is encoded by the CRYAB (also known as CRYA2, HSPB5) gene (Gene ID: 1410) in human. Alpha-crystallin B chain (CRYAB) is a small heat shock protein (sHsp) that functions as a molecular chaperone. It exhibits a characteristic beta-sheet-rich structure, enabling it to form oligomeric complexes that assist in preventing protein aggregation. It contains a conserved -crystallin structure involved in the dimerization domain, and C-terminal domain containing the IXI motif responsible for oligomer assembly. CRYAB prevents the aggregation of denatured and stress-accumulated misfolded proteins and keeps aggregation prone proteins in reservoirs of non-native, foldable intermediates within large, soluble, multimeric structures. CRYAB is particularly active under stress conditions such as oxidative stress and elevated temperatures. CRYAB is expressed higher in tissues like the lens of the eye, heart and skeletal muscles. Age-related changes in its expression have been implicated in conditions such as cataracts and cardiomyopathies. Research suggests a potential role for CRYAB in neurodegenerative diseases like Alzheimer s and malignant tumors. Mutations of the different domains are associated with myopathy, cardiomyopathy and cataract or discrete lens opacity. Application

Quality Control Testing

Evaluated by Western Blotting in mouse brain tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected -B-crystallin/CRYAB in mouse brain tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CRYAB in rat heart tissue lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CRYAB in U-2 OS cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CRYAB in human heart tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 4G15 is a ZooMAb® rabbit recombinant monoclonal antibody that detects -B-crystallin/CRYAB. It targets an epitope within 20 amino acids from the C-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
