General description
Metalloproteinase inhibitor 3 (UniProt: P35625; also known as Protein MIG-5, Tissue inhibitor of metalloproteinases 3, TIMP-3) is encoded by the TIMP3 gene (Gene ID: 7078) in human. The tissue inhibitors of metalloproteinases (TIMPs) represent a family of ubiquitous proteins that are natural inhibitors of the matrix metalloproteinases (MMPs). There are four known members of the TIMP family that are designated as TIMP-1, 2, 3, and 4. TIMPs are released during tissue damage to facilitate tissue remodeling through degradation and reorganization of the extracellular matrix. During this process MMPs are involved in inflammatory response through proteolytic maturation of cytokines. These activities are regulated through a 1:1 stoichiometric interaction of MMPs with TIMPs and these interactions are tightly controlled. TIMP3 is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to produce the mature form. It mediates a variety of processes including matrix regulation and turnover, inflammation, and angiogenesis, through reversible inhibition of MMPs. It is also reported to modulate the processing of amyloid precursor protein and apolipoprotein E receptor ApoER2 by inhibiting two -secretases (ADAM10 and ADAM17). TIMP3 also functions as a tumor suppressor and a potent inhibitor of angiogenesis. It exerts its anti-angiogenic effect by directly interacting with VEGF receptor-2/KDR, preventing its binding to the VEGFA ligand. Immunogen
Application
Evaluated by Western Blotting in mouse kidney tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TIMP3 in mouse kidney tissue lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TIMP3 in lysate from SK-MEL-28 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TIMP3 in human placenta tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound TIMP3 peptide with at least ten million-fold (10,000,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
