Cystathionine gamma-lyase (UniProt: P32929; also known as EC:4.4.1.1, CGL, CSE, Cysteine desulfhydrase, Cysteine-protein sulfhydrase, Gamma-cystathionase, Homocysteine desulfhydrase) is encoded by the CTH gene (Gene ID: 1491) in human. Cystathionine gamma-lyase (CTH) is a pyridoxal phosphate-dependent enzyme that catalyzes the conversion of cystathionine to cysteine, ammonia, and -ketobutyrate in the transsulfuration (TSS) pathway. CTH is a homotetramer, with each subunit binding to a pyridoxal phosphate cofactor essential for its enzymatic activity. CTH can utilize L-cysteine and L-homocysteine as substrates to produce the endogenous gaseous signaling molecule hydrogen sulfide (H2S) via desulfhydration of cysteine. CTH is predominantly expressed in the liver, kidney, and muscles. Dysregulation of CTH expression and activity has been implicated in numerous diseases, including cardiovascular diseases, where altered H₂S production affects vascular tone and inflammation. Mutations in the CTH gene have been associated with cystathioninuria, a metabolic disorder characterized by elevated cystathionine levels. Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CTH in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CTH in SH-SY5Y cell lysate and a 1:10,000 dilution from HepG2 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound with CTH peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CTH in human liver and rat liver tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 2F12 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects CTH. It targets an epitope within 25 amino acids from the C-terminal region.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
