General description
Antigen-presenting glycoprotein CD1d1 (UniProt: P11609; also known as CD1d.1) is encoded by the Cd1d1 (also known as Cd1.1) gene (Gene ID: 12479) in murine species. CD1d is a transmembrane glycoprotein, part of the CD1 family of 5 antigen presenting molecules, that are related to Major histocompatibility complex class I (MHC-I) and II (MHC-II) molecules. The CD1 family is subdivided into group 1 (CD1a, CD1b, CD1c and CD1e) and group 2 (CD1d) based on sequence homology. CD1d mediates the presentation of lipid and glycolipid antigens derived from self or microbial sources to natural killer T (NKT) cells, which play a crucial role in immune responses. These antigens are presented via heterodimers formed with beta-2 microglobulin. CD1d is predominantly localized to late endosomes and lysosomes, facilitated by a tyrosine-based motif in its cytoplasmic tail, which enables intracellular trafficking. The protein is involved in both innate and adaptive immunity, particularly in regulating inflammatory responses through interactions with invariant NKT cells. CD1d expression is group-enriched in the intestine, liver, and lymphoid tissues, with enhanced expression observed in specific cell types such as Kupffer cells, macrophages, Hofbauer cells, and monocytes, underscoring its role in lipid antigen presentation and immune surveillance. CD1d has been implicated in diseases such as glioblastoma, where its expression correlates with immune system activity, and it is considered a potential therapeutic target in NKT cell-based cancer immunotherapy. CD1d expression is prognostic in certain lung cancers and adenocarcinoma. Application
Evaluated by Western Blotting in RAW 264.7 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CD1d1 in RAW 264.7 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD1d1 in EL4 cell lysate and mouse thymus tissue lysate and a 1:10,000 dilution in rat thymus tissue lysate.
Affinity Binding Assay: A representative lot of this antibody bound with CD1d1 peptide with at least one million-fold (1,000,000X) higher affinity than with non-specific control peptide.
Flow Cytometry Analysis: 1 µg from a representative lot detected CD1d1 in one million C57BL6 mouse splenocytes.
Immunocytochemistry Analysis : A 1:100 dilution from a representative lot detected CD1d1 in RAW 264.7 cells.
