Interleukin-1 receptor type 1 (UniProt: P13504; also known as IL-1R-1, IL-1RT-1, IL-1RT1, CD121 antigen-like family member A, Interleukin-1 receptor alpha (IL-1R-alpha), Interleukin-1 receptor type I, p80, CD121a, mIL-1R1, mIL-1RI, sIL-1R1, sIL-1RI) is encoded by the Il1r1 (also known as Il-1r1, Il1ra) gene (Gene ID: 16177) in murine species. Interleukin-1 receptor type 1 (IL1R1) is a transmembrane glycoprotein that belongs to the interleukin-1 receptor family and the immunoglobulin superfamily. IL1R1 comprises three extracellular immunoglobulin-like domains that facilitate ligand binding, a transmembrane region, and a cytoplasmic Toll/IL-1 receptor (TIR) domain that mediates downstream signal transduction. This receptor is integral to the formation of the high-affinity interleukin-1 receptor complex by interacting with interleukin-1 receptor accessory protein (IL1RAP) with predominantly interleukin-1 alpha (IL-1 ), interleukin-1 beta (IL-1β), and the interleukin-1 receptor antagonist (IL-1Ra). IL1R1 plays a pivotal role in the immune system by mediating interleukin-1-dependent activation of key signaling pathways, including nuclear factor-kappa B (NF- B), mitogen-activated protein kinase (MAPK), and stress kinase pathways. These signaling cascades regulate inflammatory responses, apoptosis, and cell proliferation. IL1R1 is strongly expressed in immune cells such as macrophages, monocytes, and T cells, as well as in non-hematopoietic cells such as endothelial cells and fibroblasts. It is involved in the regulation of fever, acute-phase protein production, and immune responses to infections and tissue injury. Dysregulation of IL1R1-mediated pathways is associated with several pathological conditions, including autoimmune diseases (e.g., rheumatoid arthritis and systemic lupus erythematosus), inflammatory disorders, and certain cancers. Application
Quality Control Testing
Evaluated by Western Blotting in THP-1 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IL1R1 in THP-1 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected IL1R1 in lysate from human bone marrow tissue and rat liver tissue.
Affinity Binding Assay: A representative lot of this antibody bound with IL1R1 peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IL1R1 in NIH/3T3 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2J18 is a ZooMAb® rabbit recombinant monoclonal antibody that detects IL1R1. It targets an epitope within 25 amino acids from the N-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
