General description

Dynactin subunit 1 (UniProt: Q14203; also known as 150 kDa dynein-associated polypeptide, DAP-150 (DP-150), p135, p150-glued) is encoded by the DCTN1 gene (Gene ID: 1639) in human. Dynactin subunit 1 (DCTN1) is a critical component of the dynactin complex involved in intracellular transport and cytoskeletal organization. Dynactin subunit 1 contains coiled-coil domains for oligomerization, a CAP-Gly domain for microtubule binding, and a basic region that interacts with dynein motors. Dynactin subunit 1 facilitates dynein-mediated transport of organelles, vesicles, and proteins along microtubules, playing an essential role in mitosis, endocytosis, and synaptic signaling. Dynactin subunit 1 is ubiquitously expressed, with higher levels in neuronal tissues. It localizes to microtubules and the cytoplasm, where it forms complexes with dynein and other cytoskeletal proteins. It interacts with TDP-43, influencing its nuclear-cytoplasmic transport and aggregation, as well as other proteins implicated in cellular trafficking and signaling pathways. Mutations in DCTN1, such as the G59S mutation, are associated with neurodegenerative diseases, including Perry syndrome and amyotrophic lateral sclerosis (ALS), characterized by protein aggregation and disrupted axonal transport. Dysregulation of Dynactin subunit 1 contributes to cellular dysfunction, neurodegeneration, and impaired vesicle trafficking, highlighting its therapeutic potential in targeting cytoskeletal and transport-related disorders. KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human DCTN1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected DCTN1 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected DCTN1 in lysates from Neuro-2a and U-2 OS cells, and mouse brain tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected DCTN1 in human cerebral cortex tissue sections.

Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated DCTN1 in HeLa cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound with DCTN1 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2B11 is a ZooMAb® rabbit recombinant monoclonal antibody that detects DCTN1. It targets an epitope within 25 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
