General description

Desmoplakin (UniProt: P15924; also known as DP, 250/210 kDa paraneoplastic pemphigus antigen) is encoded by the DSP gene (Gene ID: 1832) in human. Desmoplakin is a crucial component of the desmosomal complex, playing a pivotal role in maintaining the structural integrity of tissues that experience mechanical stress, such as the heart, skin, and mucous membranes. Desmoplakin is a large, plakin family protein characterized by a central rod domain flanked by N-terminal and C-terminal globular domains. The N-terminal domain interacts with desmosomal cadherins, while the C-terminal domain binds to intermediate filaments, such as keratins in epithelial cells and desmin in cardiac muscle cells, thus anchoring the desmosomal plaque to the cytoskeleton. Desmoplakin is ubiquitously expressed in tissues with high mechanical demands. It is localized at the desmosomes, specialized cell junctions that confer mechanical strength by linking the cytoskeletons of adjacent cells. Mutations in the desmoplakin gene can lead to a variety of diseases, most notably arrhythmogenic right ventricular cardiomyopathy (ARVC), a condition characterized by progressive replacement of cardiac muscle with fibrous and fatty tissue, leading to arrhythmias and heart failure. Additionally, desmoplakin mutations are implicated in skin disorders such as palmoplantar keratoderma and epidermolysis bullosa. KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal half of human Desmoplakin.

Application

Quality Control Testing

Evaluated by Western Blotting in HaCaT cell lysate.

Western Blotting Analysis: A 1:20,000 dilution of this antibody detected Desmoplakin in HaCaT cell lysate.

Tested Applications

Western Blotting Analysis: A 1:50,000 dilution from a representative lot detected Desmoplakin in rat heart tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Desmoplakin in A-431 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Desmoplakin in human heart tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with Desmoplakin peptide with at least one hundred million-fold (100,000,000X) higher affinity than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Desmoplakin in A-431 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 3M12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Desmoplakin. It targets an epitope within 22 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
