General description
POU domain, class 3, transcription factor 2 (UniProt: P20265; also known as Brain-specific homeobox/POU domain protein 2 (Brain-2; Brn-2), Nervous system-specific octamer-binding transcription factor N-Oct-3, Octamer-binding protein 7 (Oct-7), Octamer-binding transcription factor 7 (OTF-7)) is encoded by the POU3F2 (also known as BRN2, OCT7, OTF7) gene (Gene ID: 5454) in human. The POU domain, class 3, transcription factor 2 (POU3F2), also known as Brn-2, is a member of the POU-domain family of transcription factors, characterized by a bipartite DNA-binding domain comprising the POU-specific (POUS) and POU-homeodomain (POUH) regions. POU3F2 plays a critical role in neurogenesis and neuronal differentiation, particularly in the development of the hypothalamus and cortex. It binds to a specific octamer DNA sequence (5′-ATGCAT-3′) and regulates the expression of genes involved in neural development. POU3F2 is predominantly localized in the nucleus and is highly expressed in neural tissues, particularly in progenitor cells and differentiated neurons. It interacts with other neural transcription factors, such as SOX11, SOX4, and SOX2, to synergistically regulate target gene expression. POU3F2 is involved in several key pathways, including the Notch signaling cascade and the regulation of corticotropin-releasing hormone (CRH) genes, which are essential for stress response and hypothalamic function. Mutations or dysregulation of POU3F2 have been implicated in neurodevelopmental disorders and glioblastoma, where it contributes to tumor progression by promoting neural stem-like properties. Its role in epithelial-mesenchymal transition (EMT) and cancer stem cell (CSC) maintenance has also been noted, linking it to metastasis and therapy resistance in cancers. Evaluated by Western Blotting in SK-N-MC cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected POU3F2 in SK-N-MC cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected POU3F2 in human brain tissue, mouse brain tissue, and rat brain tissue lysates.
Immunocytochemistry Analysis: A 1:1,000 Dilution from a representative lot detected POU3F2 in U-251 MG cells.
Affinity Binding Assay: A representative lot of this antibody bound with POU3F2 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected POU3F2 in mouse brain tissue sections.
