General description
Exportin-1 (UniProt O14980; also known as Chromosome region maintenance 1 protein homolog, CRM1 homolog, Exp1) is encoded by the XPO1 (also known as CRM1) gene (Gene ID 7514) in human. It is a nuclear export protein that mediates leucine-rich nuclear export signal (NES)-dependent protein transport. Most nuclear importins & exportins, including exportin-1, belong to the beta-karyopherin (KAP) family of proteins, Importins bind their cargoes with either nuclear localization signal (NLS) or M9 domain-containing hnRNP proteins in the cytoplasm, while exportins target their substrates via their nuclear export signal (NES) or M9 domain inside the nucleus. Structurally, XPO1 comprises multiple HEAT repeats that facilitate its interaction with cargo proteins bearing leucine-rich nuclear export signals (NES). Its expression is ubiquitous but is notably elevated in various malignancies, including hematological cancers and solid tumors. Exportin-1 is predominantly present in nucleus but shuttles between the nucleus/nucleolus and the cytoplasm. In the nucleus, Exportin-1 forms a complex with its cargo and the GTPase Ran in its active GTP-bound form, enabling efficient docking at the nuclear pore complex for transport. Exportin-1 plays a significant role in maintaining cellular homeostasis and regulating key processes such as the cell cycle and apoptosis. The upregulation and mutation of XPO1, such as the E571K variant, have been implicated in the pathogenesis of several cancers, making it a promising target for therapeutic intervention. Inhibitors of XPO1, such as selinexor, have shown efficacy in clinical trials, providing a novel strategy for cancer treatment by re-establishing the nuclear localization of tumor suppressor proteins and enhancing anti-tumor immunity. Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Exportin-1/CRM1 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Exportin-1/CRM1 in lysate from MCF7 cells.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Exportin-1/CRM1 in HeLa cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Exportin-1/CRM1 in human testis tissue sections.

Biochem/physiol Actions
Clone 1B2 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects Exportin-1/CRM1. It targets an epitope within 18 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
