General description
Transcription factor Maf (UniProt: O75444; also known as Transcription factor Maf, Proto-oncogene c-Maf, V-maf musculoaponeurotic fibrosarcoma oncogene homolog) is encoded by the MAF gene (Gene ID: 4094) in human. The c-Maf transcription factor is part of the basic leucine zipper (bZIP) family of proteins, characterized by a basic leucine zipper (bZip) structure that facilitates dimerization and DNA-binding. It plays a pivotal role in cellular differentiation, development, and homeostasis. It forms a homodimer or a heterodimer with other bHLH-Zip transcription factors. c-Maf functions as both a transcriptional activator and repressor, depending on its binding partners and context. c-Maf is critical in processes such as lens formation, osteoblast differentiation, macrophage function, and insulin transcription in pancreatic β-cells. It binds to Maf recognition elements (MAREs) via its bZIP domain, regulating downstream genes. Mutations in c-MAF gene are linked to congenital cataracts, Ayme-Gripp syndrome, and Duane retraction syndrome, among other conditions. Additionally, c-MAF has been implicated in oncogenesis, particularly in multiple myeloma, through its role in dysregulated gene expression pathways. Its activity is modulated by phosphorylation and interactions with other transcription factors, such as MYB, affecting immunity and apoptosis in T cells. Application
Evaluated by Western Blotting in RPMI 8226 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected c-Maf in RPMI 8226 cell lysate.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound with c-Maf peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated c-Maf from RPMI 8226 cell lysate.
Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.
