General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Killer cell lectin-like receptor subfamily B member 1A (UniProt: P27811; also known as NKR-P1A, CD161 antigen-like family member A, Lymphocyte antigen 55A (Ly-55A), NKR-P1.7, Natural killer cell surface protein P1-2 (NKR-P1 2), CD161a) is encoded by the Klrb1a (also known as Ly55, Ly55a, Nkrp1a) gene (Gene ID: 17057) in murine species. The Killer cell lectin-like receptor subfamily B member 1 (KLRB1), also referred to as CD161, is a type II transmembrane protein. It is a member of the C-type lectin superfamily and plays a critical role in immune cell regulation, particularly in natural killer (NK) cells and subsets of T cells. KLRB1 has an extracellular C-type lectin domain responsible for ligand binding, a transmembrane domain, and a cytoplasmic tail. Its primary ligand is LLT1, which modulates immune responses by inhibiting NK cell cytotoxic activity. KLRB1 is expressed predominantly in immune cells, including NK cells, CD4+ T cells, and CD8+ T cells. It has been implicated in inhibiting NK cell-mediated cytotoxicity and promoting cytokine release, thereby contributing to immune homeostasis. Dysregulation of KLRB1 expression has been associated with autoimmune disorders such as multiple sclerosis and rheumatoid arthritis, as well as poor prognosis in cancers, including glioblastoma and hepatocellular carcinoma. Additionally, KLRB1 expression is altered in conditions like inflammatory bowel disease and atopic dermatitis. Furthermore, KLRB1′s role extends to inflammatory signaling pathways, likely influencing the immune microenvironment in cancer and infections. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the extracellular domain of human KLRB1.

Application

Quality Control Testing
Evaluated by Western Blotting in His-tagged, recombinant fragment of mouse KLRB1.

Western Blotting Analysis: A 1:50,000 dilution of this antibody detected His-tagged, recombinant fragment of mouse KLRB1.
Tested Applications
Immunocytochemistry Analysis : A 1:100 Dilution from a representative lot detected KLRB1A in HEK293T cells transfected with Klrb1a-GFP plasmid.

Affinity Binding Assay: A representative lot of this antibody bound with KLRB1A peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2B12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects KLRB1A/1B/1C. It targets an epitope within 20 amino acids from the extracellular domain.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
