General description

UDP-glucuronosyltransferase 2B7 (UniProt: P16662; also known as UDPGT 2B7, UGT2B7, 3,4-catechol estrogen-specific UDPGT, UDP-glucuronosyltransferase 2B9, UDPGT 2B9, UDPGTh-2, EC:2.4.1.17) is encoded by the UGT2B7 (also known as UGTB2B9) gene (Gene ID: 7364) in human. The UGTs serve a major role in the conjugation and subsequent elimination of potentially toxic xenobiotics and endogenous compounds. UGT2B7 has unique specificity for 3,4-catechol estrogens and estriol, suggesting that it may play an important role in regulating the level and activity of these potent estrogen metabolites. It is also the major isoform for the metabolism of morphine to the main metabolites. UGT2B7 is a phase II metabolism isoenzyme active in the liver, kidneys, gastrointestinal tract and brain. Structurally, UGT2B7 is a single-pass membrane protein, synthesized with a signal peptide (aa 1-23), which is cleaved off to produce a mature form (aa 24-529) that contains a small transmembrane domain (aa 493-509). It is localized predominantly in the endoplasmic reticulum and nuclear membranes of cells. Recent studies have highlighted the complexity of its expression profile, revealing that UGT2B7 expression is regulated by various transcription factors, including p53, which can enhance its transcription in response to cytotoxic stress, thereby influencing drug metabolism and susceptibility to drug-induced toxicity. Additionally, UGT2B7 genetic variants have been associated with altered drug metabolism, impacting therapeutic efficacy and toxicity, particularly in cancer treatments. Research study indicated that UGT2B7 plays a role in renal cell carcinoma (RCC) progression and could be a potential prognostic biomarker for patients with RCC. Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected UGT2B7 in HepG2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected UGT2B7 in lysates from human liver tissue, human kidney tissue, and mouse kidney tissue, and a 1:10,000 dilution from a representative lot detected UGT2B7 in lysate from rat kidney tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected UGT2B7 in human kidney tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected UGT2B7 in HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound UGT2B7 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 4C11 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects UGT2B7. It targets an epitope within 22 amino acids from the internal region. It may cross-react to other family members of UGT2B and UGT2A due to high protein sequence homology.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
