General description

T-cell antigen CD7 (UniProt: P09564; also known as GP40, T-cell leukemia antigen, T-cell surface antigen Leu-9, TP41) is encoded by the CD7 gene (Gene ID: 924) in human. CD7 is a transmembrane glycoprotein expressed predominantly on T cells, natural killer (NK) cells, and their precursors. It belongs to the immunoglobulin superfamily and functions as a single-pass type I membrane protein. CD7 plays a critical role in immune regulation by acting as a costimulatory molecule during T-cell activation, particularly upon binding to its ligand, SECTM1 (secreted and transmembrane protein 1). This interaction facilitates the production of cytokines like interleukin-2 (IL-2) and interferon-gamma (IFN- ), enhancing immune responses. CD7 is involved in intracellular signaling pathways, including the phosphatidylinositol 3-kinase (PI3K) pathway, which is crucial for cellular adhesion and migration. Its expression is tightly regulated by transcriptional and post-transcriptional mechanisms, and it is upregulated in certain immune activation states. CD7 interacts with other proteins such as PI3K and SECTM1, influencing processes like monocyte migration and T-cell infiltration into tumors. Dysregulation of CD7 has been implicated in various diseases, including lymphomas, autoimmune disorders like systemic sclerosis, and immune deficiencies. Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the internal region of human CD7.

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CD7 in Jurkat cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CD7 in human peripheral blood mononuclear cells (PBMC) and human thymus tissue lysates.

Affinity Binding Assay: A representative lot of this antibody bound with CD7 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD7 in HepG2 cells and Jurkat cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected CD7 in one million human peripheral blood mononuclear cells (PBMC).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1E19 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CD7. It targets an epitope within 25 amino acids from the internal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

0.3 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
