General description
RecQ-like DNA helicase BLM (UniProt: P54132; also known as EC:5.6.2.4, Bloom syndrome protein, DNA 3′-5′ helicase BLM, DNA helicase, RecQ-like type 2 (RecQ2), RecQ protein-like 3) is encoded by the BLM (also known as RECQ2, RECQL3) gene (Gene ID: 641) in human. The BLM protein is a highly conserved member of the RecQ helicase family, characterized by its 3′ to 5′ ATP-dependent DNA unwinding activity. This helicase plays a vital role in maintaining genomic stability by participating in DNA replication, repair, and homologous recombination (HR). BLM is integral to several DNA repair pathways, including the resolution of double-strand breaks (DSBs) via HR, synthesis-dependent strand annealing (SDSA), and the dissolution of double Holliday junctions. Structurally, it contains conserved helicase domains, a RecQ C-terminal (RQC) domain for protein-protein interactions, and a helicase-RNase D C-terminal (HRDC) domain that facilitates DNA binding. BLM predominantly localizes to the nucleus, where it colocalizes with DNA repair markers such as γH2AX, RAD51, and 53BP1 during the S and G2 phases of the cell cycle. Additionally, it interacts with topoisomerase 3α (TOP3A) and RecQ-mediated genome instability proteins (RMI1 and RMI2) to form a complex vital for resolving recombination intermediates. Post-translational modifications, including phosphorylation and ubiquitination, regulate its activity and localization. Mutations in the BLM gene result in Bloom syndrome, a rare autosomal recessive disorder characterized by chromosomal instability, growth retardation, immunodeficiency, and a predisposition to multiple cancers. Evaluated by Western Blotting in rat brain tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected BLM in rat brain tissue lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected BLM in nuclear extract from HEK-293 cells and a 1:1,000 dilution from HeLa and NIH/3T3 cell lysates.
Flow Cytometry Analysis: 1 µg from a representative lot detected BLM in one million HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound with BLM peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
