General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Calpain-2 catalytic subunit (UniProt: P17655 ; also known as EC:3.4.22.53, Calcium-activated neutral proteinase 2 (CANP 2), Calpain M-type, Calpain large polypeptide L2 Calpain-2 large subunit,Millimolar-calpain (M-calpain)) is encoded by the CAPN2 (also known as CANPL2) gene (Gene ID: 824) in human. Calpain-2, a catalytic subunit of the ubiquitous calpain system, belongs to the family of calcium-dependent, non-lysosomal cysteine proteases. It forms a heterodimer with a common regulatory subunit (calpain small subunit 1, CSS1). The calpain-2 catalytic subunit consists of four domains: a protease core domain, a regulatory domain, a calcium-binding domain, and a C2-like domain that facilitates membrane interactions. Calpain-2 is predominantly cytoplasmic, but upon activation by calcium ions, it translocates to the plasma membrane where it interacts with its substrates. It can also translocate to the nucleus and nuclear membrane upon increased intracellular levels, contributing to apoptosis, nuclear remodeling, and protein degradation. It facilitates the cleavage of nuclear substrates like DNA topoisomerase I, contributes to nuclear envelope breakdown, and can be found in the nucleoli of tumor cells. This protease is broadly expressed in a variety of tissues, with particularly high levels in the brain, skeletal muscles, and the cardiovascular system. Calpain-2 is involved in numerous physiological processes, including cytoskeletal remodeling, cell motility, signal transduction, cell proliferation, and apoptosis. It regulates these processes by cleaving specific substrates, such as cytoskeletal proteins (e.g., talin, spectrin), transcription factors, and signaling molecules. Calpain-2 is critically involved in pathways like focal adhesion dynamics, integrin signaling, and the regulation of inflammatory responses. Dysregulation of calpain-2 activity is implicated in various pathological conditions. Overactivation contributes to neurodegenerative disorders such as Alzheimer′s disease and Parkinson′s disease, where it mediates proteolysis of key neuronal proteins. It is also associated with ischemic injury, cancer progression, and cardiovascular diseases, where it promotes cell migration, invasion, and matrix degradation. Conversely, insufficient calpain-2 function can impair cellular repair mechanisms. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Application

Quality Control Testing
Evaluated by Western Blotting in HEK-293 cell lysate. 

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Calpain 2-CAPN2 in HEK-293 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Calpain 2 in SK-BR-3 and ACHN cell lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Calpain 2 in human kidney tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected Calpain 2 in SK-BR-3 cells.
Affinity Binding Assay: A representative lot of this antibody bound with Calpain 2 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 3H4 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Calpain 2. It targets an epitope within 21 amino acids from the internal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
