General description

CD209 antigen (UniProt: Q9NNX6; also known as C-type lectin domain family 4 member L, Dendritic cell-specific ICAM-3-grabbing non-integrin 1 (DC-SIGN; DC-SIGN1)) is encoded by the CD209 (also known as CLEC4L) gene (Gene ID: 30835) in human. CD209 is a C-type lectin receptor primarily expressed on dendritic cells and macrophages. CD209 consists of an extracellular domain containing a carbohydrate recognition domain (CRD), a neck region that mediates tetramerization, a transmembrane domain, and a cytoplasmic tail essential for intracellular signaling. CD209 plays a critical role in pathogen recognition and immune modulation by binding high-mannose glycans present on pathogens such as HIV, SARS-CoV-2, and Mycobacterium tuberculosis. This interaction facilitates pathogen uptake through endocytosis and phagocytosis, triggering downstream signaling pathways that regulate innate and adaptive immunity. CD209 is predominantly localized to the plasma membrane and endosomal compartments, where it mediates antigen presentation and cytokine production. It interacts with intracellular proteins such as LSP1 and other signaling molecules to modulate immune responses. Dysregulation of CD209 is implicated in various diseases, including infectious diseases, autoimmune disorders, and cancer, where altered expression or function contributes to immune evasion and chronic inflammation. Immunogen

KLH-conjugated linear peptide corresponding to amino acids from the C-terminal extracellular domain of human CD209.

Application

Quality Control Testing

Evaluated by Western Blotting in LoVo cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD209 in LoVo cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound with CD209 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD209 in human colon and human liver tissue sections.

Flow Cytometry Analysis: 1 µg from a representative lot detected CD209 in Human Peripheral Blood Monocytes Derived Mature Dendritic Cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD209/DC-SIGN in THP-1 cells .

Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 4B5 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CD209. It targets an epitope within 16 amino acids from the C-terminal extracellular domain.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
