Ubiquitin-like protein ISG15 (UniProt: P05161; also known as Interferon-induced 15 kDa protein, Interferon-induced 17 kDa protein (IP17), Ubiquitin cross-reactive protein (hUCRP)) is encoded by the ISG15 (also known as G1P2, UCRP) gene (Gene ID: 9636) in human. Interferon-stimulated gene product 15 (ISG15) is a ubiquitin-like modifier protein encoded by the ISG15 gene, which plays multifaceted roles in the innate immune system. ISG15 consists of two ubiquitin-like domains connected by a linker region, allowing it to interact with various substrates and signaling molecules. Its primary role involves conjugation to intracellular proteins via ISGylation, a process catalyzed by E1 (UBE1L), E2 (UbcH8), and E3 ligases, such as HERC5. This conjugation regulates the stability, localization, and activity of target proteins. ISG15 also acts as an extracellular cytokine, modulating immune responses by inducing interferon- (IFN- ) secretion from natural killer cells and influencing chemotactic activity toward neutrophils. ISG15 is strongly upregulated in response to type I interferons (IFN- /β) during viral and microbial infections. It inhibits viral replication and assembly by targeting both viral and cellular proteins, thereby serving an antiviral function., ISG15 is localized in the cytoplasm and nucleus, and unconjugated (free) ISG15 can act as a signaling molecule. Its expression profile reveals tissue enrichment in immune-related cells, including macrophages and dendritic cells, and elevated levels in infected or inflamed tissues. Dysregulation of ISG15 has been implicated in autoimmune diseases such as systemic lupus erythematosus (SLE), where excessive ISG15 activity can lead to prolonged interferon responses. ISG15 is associated with multiple diseases, including immunodeficiency 38 with basal ganglia calcification, influenza, and SARS-CoV-2 infection. Recent studies have highlighted ISG15′s interaction with DNA repair pathways, linking it to genome stability and replication processes. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Application

Quality Control Testing
Evaluated by Western Blotting in lysate from HeLa cells treated with IFN- and Calyculin A.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected ISG15 in lysate from HeLa cells treated with IFN- (1000IU/mL; 18 h) and Calyculin A: 0.1 M, 15 min.).
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ISG15 in lysate from human peripheral blood mononuclear cells (PBMC) and human liver tissue.
Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated ISG15 in HeLa cell lysate treated with IFN- and Calyculin A.

Affinity Binding Assay: A representative lot of this antibody bound with ISG15 peptide with at least one billion-fold (1,000,000,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

General description

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human ISG15.

Other Notes

Clone 3L15 is a ZooMAb® rabbit recombinant monoclonal antibody that detects ISG15. It targets an epitope within 25 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
