General description
GTP-binding nuclear protein Ran (UniProt: P62826; also known as Androgen receptor-associated protein 24, GTPase Ran, Ras-like protein TC4, Ras-related nuclear protein) is encoded by the RAN (also known as ARA24, OK/SW-cl.81) gene (Gene ID: 5901) in human. Ran is a small GTP binding protein of the RAS superfamily that is predominantly expressed in the nucleus during interphase and is dispersed throughout the cytoplasm during mitosis. The GTP-bound Ran (RanGTP) is considered active, while the GDP-bound Ran (RanGDP) is inactive. These two alternate states are regulated by two classes of partner proteins that either promote the RanGTP or the RanGDP form. RanGEF (guanine-nucleotide exchange factor, RCC1) enhances the formation of RanGTP and RanGAP (GTPase-activating protein) and RanBPs (Ran-binding proteins: RanBP1 and RanBP2), which increase the GTPase activity of Ran promote the formation of RanGDP. The GTPase activity of Ran is involved in nucleocytoplasmic transport, participating both to the import and the export of proteins and RNAs from the nucleus. Ran is shown to control cargo loading and release by transport receptors, such as importin , in the proper compartment, which ensures the directionality of the transport. Its interaction with RANBP1 induces a conformation change in the complex formed by XPO1 and RAN that triggers the release of the nuclear export signal of cargo proteins. In its GTP-bound form RAN also triggers microtubule assembly at mitotic chromosomes and is required for normal mitotic spindle assembly and chromosome segregation. Application
Evaluated by Western Blotting in Jurkat cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected RAN in Jurkat cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected RAN in lysates from NIH/3T3 cells, A-431 cells, and rat kidney tissue.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected RAN in human colon tissue sections.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected RAN in PC-3 cells.
Affinity Binding Assay: A representative lot of this antibody bound RAN peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
