General description

Paired box protein Pax-5 (UniProt: Q02548; also known as B-cell-specific transcription factor, BSAP) is encoded by the PAX5 gene (Gene ID: 5079) in human. PAX5 protein is a critical transcription factor belonging to the paired box (PAX) family, plays a pivotal role in B-cell lineage commitment and differentiation. It fulfills a dual role by repressing B-lineage inappropriate genes and simultaneously activating B-lineage-specific genes thus effectively orchestrating the developmental transition from progenitor cells to mature B cells. Structurally, PAX5 is characterized by a paired DNA binding domain (aa 16-142) that enables it to bind DNA thus regulating the expression of genes essential for B-cell development. Its expression is predominantly nuclear and is expressed at early B-cell differentiation, in the developing CNS and in adult testis. Eleven isoforms of PAX5 have been described that are produced by alternative splicing. Mutations in PAX genes are known to be associated with numerous human diseases (e.g., microphthalmia, anophthalmia, coloboma, hypothyroidism, acute lymphoblastic leukemia). PAX5 is notably involved in various hematological malignancies, particularly acute lymphoblastic leukemia (ALL), where chromosomal aberrations and mutations lead to either loss of function or the formation of fusion proteins that disrupt normal transcriptional regulation. This dysregulation in PAX5 gene contributes to leukemogenesis, highlighting PAX5 as a potential biomarker and therapeutic target in B-cell malignancies. KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal region of human PAX5.

Application

Quality Control Testing

Evaluated by Western Blotting in Ramos cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected PAX5 in Ramos cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PAX5 in lysate from Raji cells.

Affinity Binding Assay: A representative lot of this antibody bound PAX5 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PAX5 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PAX5 in Ramos cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1B14 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PAX5. It targets an epitope within 20 amino acids from the C-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
