General description

Microtubule-associated protein RP/EB family member 1 (UniProt: Q15691; also known as APC-binding protein EB1, End-binding protein 1) is encoded by the MAPRE1 gene (Gene ID: 22919) in human. Microtubule-associated protein RP/EB family member 1 (EB1/MAPRE1) is a microtubule plus end-tracking protein (+TIP). It harbors an EB1 homology domain (aa 185-255) that contains a highly conserved hydrophobic cavity and through the SxIP motif EB1/MAPRE1 binds to +TIPs and autonomously tracks microtubule tips independent of any binding partners. EB1/MAPRE1 binds to an array of structurally and functionally unrelated +TIP binding partners, including APC tumor suppressor protein, MACF, CLIP170, CLIP-associated proteins (CLASPs), and MCAK among others. EB1/MAPRE1 involved in a variety of cellular processes, including establishment and maintenance of cell polarity, search and capture of chromosomes during mitosis, and positioning of the mitotic spindle during asymmetric cell division. It is vital for cell cycle regulation and mitotic progression, ensuring genome stability. It is ubiquitously expressed across various tissues, with higher expression levels noted in cancerous tissues such as hepatocellular carcinoma and colorectal cancer. EB1/MAPRE1 is implicated in cancer progression, enhances in vitro tumorigenesis, and associates with poor prognosis of hepatocellular carcinoma and breast cancer. Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human EB1/MAPRE1.

Application

Quality Control Testing

Evaluated by Western Blotting in MOLT-4 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected EB1/MAPRE1 in MOLT-4 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected EB1/MAPRE1 in lysates from rat testis tissue and NIH/3T3 cells.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected EB1/MAPRE1 in HepG2 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected EB1/MAPRE1 in NIH/3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound with EB1/MAPRE1 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1M14 is a ZooMAb® rabbit recombinant monoclonal antibody that detects EB1/MAPRE1. It targets an epitope within 17 amino acids from the C-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
