General description
Natural resistance-associated macrophage protein 2 (UniProt: P49281; also known as NRAMP 2, Divalent cation transporter 1, Divalent metal transporter 1, DMT-1, Solute carrier family 11 member 2) is encoded by the SLC11A2 (also known as DCT1, DMT1, NRAMP2) gene (Gene ID: 4891) in human. It is a proton-coupled Fe2+ transporter that is essential for iron uptake in enterocytes and for transferrin-associated endosomal iron transport in many other cell types. SLC11A2 can transport manganese, cobalt, cadmium, nickel, and vanadium. Structurally, SLC11A2 is a multi-pass membrane protein characterized by multiple transmembrane domains that allow it to function as a symporter, coupling the transport of protons with divalent cations such as Fe²⁺. It is ubiquitously expressed and also found in erythroid progenitors. Its expression is particularly prominent in the duodenum, where it is essential for the absorption of dietary iron, and it is also found in the brain, liver, and various immune cells. SLC11A2 is localized to the plasma membrane and endosomal compartments, where it mediates the uptake of iron in a pH-dependent manner. Five isoforms of SLC11A2 have been described that are produced by alternative splicing. have been described that are produced by alternative splicing. Mutations in the SLC11A2 gene have been implicated in several diseases, most notably hypochromic microcytic anemia and iron overload disorders, as these mutations can disrupt normal iron uptake and lead to abnormal iron accumulation in tissues. Additionally, SLC11A2 dysfuntion has been associated with the pathogenesis of neurodegenerative diseases, such as Alzheimer′s disease, due to its role in regulating metal ion homeostasis that influences amyloid beta aggregation. Recent study highlights pivotal role of SLC11A1 and SLC11A2 in iron metabolism, cellular homeostasis, processes linked to oncogenesis and, suggest their potential as biomarkers and therapeutic targets. Application
Evaluated by Western Blotting in HepG2 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SLC11A2 in HepG2 cell lysate.
Tested Applications
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected SLC11A2 in HepG2 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SLC11A2 in human cerebral cortex tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound SLC11A2 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
