S-乙酰巯基丁二酸酐是一种胺反应性试剂，含有巯基。当被胺亲核试剂攻击时，酸酐区域打开，形成胺键。但是，在该开环反应过程中，形成了游离的羧酸根基团，使分子带负电。电荷逆转影响蛋白质的活性和分子的构象。
