PSI²

PSI² is a stable and sustainably prepared P(V) reagent developed by the Baran Lab for the synthesis of phosphorodithioate oligonucleotides. The phosphorodithioate linkages afforded by PSI² are effectively cleaved by RNAse-H while remaining stable to nucleases, and can be formed rapidly in high yield. The synthetic methodology accessed by PSI² and other PSI reagents (see ALD00602 and ALD00604) allows at will installation of a greater diversity of linkages to DNA and other nucleotides with comparable performance to existing state-of-the-art P(III) based reagents. The resulting compounds have applications in gene-based therapeutics such as antisense oligonucleotides (ASOs).
