Biochem/physiol Actions

DHPC可保留溶解蛋白的天然构象及活性。蛋白质的三维结构和活性不仅能够在通常的最大溶解浓度（10-15mM DHPC）下，而且在大范围的DHPC浓度（最高40mM）下均可得以保留。不仅是三维结构，而且大多数蛋白质的活性都能够在过量的DHPC中得以保留，而且这些蛋白质在DHPC中人仍能保持稳定。该结果表明DHPC与完整的膜蛋白的直接相互作用较弱。如果DHPC与膜蛋白的相互作用确实较弱，则可以保证固有的膜脂质将保持与膜蛋白的结合。DHPC的增溶机理被认为是对邻近的脂质起楔形作用，这主要是由于其庞大的极性基团和短的烃链结构。这会在相对较低的DHPC浓度下导致膜不稳定。所有研究的膜组分均以相同的DHPC/脂质比率发生溶解。该发现被认为是DHPC主要与脂质双层相互作用而不与膜蛋白相互作用的证据。其可以保留天然蛋白质结构的基本原理是DHPC无法置换完整膜蛋白中的内在膜脂质。DHPC的优点包括：DHPC可保留溶解的膜蛋白的活性。DHPC不容易被氧化，并且在很宽的pH范围（4-10）内保持稳定。当分散在水中时，DHPC形成胶束而不是脂双层。（CMC = 1.4mM）。DHPC具有很大的尺寸分布，具体取决于悬浮液中的NaCl浓度。DHPC不会干扰分光光度法的测定。
