DOPE-RGD Lipid

DOPE is a phospholipid widely used to formulate liposomes and other lipid-based nanoparticles. Liposomes are spherical vesicles made of a lipid bilayer that can encapsulate drugs or other molecules of interest. DOPE is often included because it can improve liposome stability and strengthen key structural features. RGD refers to the arginine–glycine–aspartic acid motif, a short peptide sequence that binds with high affinity to integrin receptors on cell surfaces. Integrins are cell-adhesion receptors involved in signaling, attachment, and migration. Because RGD can engage integrins, it is commonly used as a targeting ligand to help direct therapeutics toward specific cells or tissues, including tumors. Studies indicate that RGD-functionalized lipids can support targeted mRNA and drug delivery. Compared with some other delivery approaches, RGD-containing peptides may offer benefits such as greater specificity, better cellular uptake, lower immunogenicity, flexible design options, and compatibility with combination strategies. Lipid nanoparticles formulated with components such as DOPE, cholesterol, and PEGylated lipids have also been reported to improve gene-editing efficiency. Overall, RGD-bearing lipids represent a promising route to more precise mRNA delivery and gene editing. Adding targeted ligands like RGD could enhance the accuracy and effectiveness of gene therapies and enable new treatments for a range of genetic disorders.
