General description

JAK2（Janus 激酶 2）属于新兴的非受体 Janus 酪氨酸激酶家族，可调节淋巴造血系统中活化细胞因子受体下游的一系列细胞功能。免疫刺激物（例如干扰素和细胞因子）诱导 Stat 转录因子募集到与细胞因子受体相关的 JAK2 上。然后，JAK2 使近端 Stat 因子磷酸化，随后将其二聚化并转运至细胞核，并与靶基因启动子上游的 CIS 元件结合，从而调节转录。经典的 JAK/Stat 途径通过刺激病原体的增殖、分化、存活和宿主抗性，是维持正常免疫系统不可或缺的一部分。改变 JAK/Stat 信号传导以减少细胞因子诱导的促炎反应代表了抗炎治疗的有吸引力的靶标。
