Bmi1蛋白是多梳家族（PcG）蛋白的成员。PcG蛋白可形成多蛋白复合物，起转录抑制因子的作用。
几条证据已表明Bmi1可参与肿瘤发生。首先，Bmi1可与c-Myc合作在双转基因小鼠中产生淋巴瘤。其次，Bmi1可阻止衰老并使小鼠胚胎成纤维细胞永生（尽管不是人类成纤维细胞），并且与激活的H-ras基因结合可导致肿瘤转化。第三，在某些套细胞淋巴瘤中观察到BMI1基因的扩增，并在非小细胞肺癌、结肠直肠癌、多发性骨髓瘤和髓母细胞瘤的亚类中过表达，而在胶质母细胞瘤中则未发现。第四，通过致癌融合蛋白E2a-Pbx1转化人类造血祖细胞需要Bmi1。实际上，基于在野生型和bmi1缺陷型遗传背景下转移性前列腺癌的小鼠模型中差异表达的基因清单，最近发现了可以预测多种癌症治疗和生存反应的特征基因。
