General description
Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone H3 variants (H3.1, H3.2 and H3.3) have been implicated in the epigenetic memory of cellular state. Genome-wide patterns of H3 are dependent on amino acid sequence and change with cellular differentiation at developmentally regulated loci.

Application
Anti-dimethyl Histone H3 (Lys27) Antibody, Cat. No. 07-421-I, is a highly specific chicken polyclonal antibody that targets histone H3 Lys27 dimethylation (H3K27me2) and has been tested in Dot Blot, Chromatin Immunoprecipitation, and Western Blotting.
Chromatin Immunoprecipitation Analysis: 12 µL from a representative lot detected Lys27-dimethylated histone H3 occupancy at the -globin promoter region using HeLa nuclear extract (0.1 million cell eq. per ChIP) and Magna ChIP HiSens Chromatin Immunoprecipitation Kit (Cat. No. 17-10460).
Dot Blot Analysis: A 1:500 dilution from a representative lot specifically detected histone H3 peptide with K27me2 modification, but not H3 peptides with unmodified K27 and other K27 modifications, H2A, H2B, or H4 peptides by dot blot analysis using AbSurance H2A, H2B, H4 Antibody Specificity Array (Cat. No. 16-665) and AbSurance H3 Antibody Specificity Array (Cat. No. 16-667).
