General description

Cyclin E1 functions as a regulator of the cell cycle, promoting the G1 to S phase transition by binding to and activating the cyclin dependent kinase CDK2. The abundance of cyclin E1 fluctuates during a normal cell cycle and is controlled at the level of gene transcription and by ubiquitin dependent proteolysis. Non-complexed cyclin E1 is degraded by the Cul3 pathway, however when cyclin E1 is bound to CDK2 its ubiqutination is dependent on a positive phosphorylation status, which enables its recognition by SCFFbw7. The impact of cyclin E1 quantities on cell cycle regulation is reflected in the elevated levels expressed by many types of human tumours.
N-terminal c-Myc, 6His-tagged, recombinant human Smad7 full length, expressed by baculovirus in Sf21 insect cells.

Quantitative assays for investigating ubiquitination pathways are essential for advancing ubiquitin-related drug discovery research. To illustrate the utility of in vitro ubiquitination enzyme cascades, we quantify the ubiquitination of Smac/DIABLO by the E3 ligase XIAP using a functional enzymatic reaction coupled to multiple assay readouts. We compare different detection methods that can be used to test small molecules for their potential to modulate XIAP-mediated ubiquitination in the context of this assay. The data provides a basis for use of these recombinant E3 ligase cascades in various applications, including screening campaigns.
