General description
一种对称的双环化合物,可拮抗CXCL12(SDF1)与CXCR4的结合(使用大鼠和人CXCR4的IC50分别为108和245 nM),并抑制SDF1诱导的Ca2+通量(在SUP-T1和THP-1培养物中以100 ng/ml的浓度抑制100%)以及CXCR4-而非CCR5-介导的HIV感染(IC50 ≤200 nM),同时增强SDF1与CXCR7的结合(在表达CXCR7的HEK293T细胞在1 mM时约60%)和SDF1诱导β-抑制蛋白募集到CXCR7(在存在和不存在10 µM AMD3100的情况下,EC50分别为6.48和11.8 nM)。据报道,通过静脉内输液施用AMD3100可导致人、狗和小鼠体内造血干细胞动员。对趋化因子通过CXCR1/2/3或CCR1/2/3/4/5/6/7/8/9诱导的信号传导无抑制作用。也可以50 mM的H2O溶液(目录号239825)提供。
