General description
一种二苯并呋喃基磺酰胺基缬氨酸化合物，可作为有效的活性位点锌靶向MMP-12选择性抑制剂（对人MMP-12的IC50 =2 nM），对小鼠/大鼠/绵羊物种的MMP-12（分别为IC50=160、320和22.3 nM）或人MMP-3/13/14（分别为IC50=351、120和1100 nM）的活性弱得多，并且对人Agg-1/2（IC50 >5 µM），TACE（IC50 >25 µM）和MMP-1/7（IC50 >6 µM）几乎无效。MMP408在小鼠中具有口服活性，并显示可有效减轻rhMMP-12诱导的小鼠体内肺组织炎症反应超过50%（5 mg/kg，p.o.，b.i.d.）。
