General description

NF-κB (nuclear factor kappa B) is a transcription factor that plays a regulatory role in several physiological and pathological processes. The NF-κB pathway can be activated by numerous stimuli, including inflammatory cytokines, virus, bacteria, UV radiation, genotoxic stress, DNA damage, oxidative stress, etc. The IkBα/NF-κB complex is tightly regulated by both phosphorylation and ubiquitination. Phosphorylation and ubiquitination of IκBα leads to its degradation and subsequent release of phosphorylated NF-κB to the nucleus. Greater than 100 target genes, including cytokines, cyclins, p21, Bcl-2, and c-Myc are known to be transcribed by NF-kB.
The NFκB pathway plays a central role in regulating immune response, inflammation, cell survival, and proliferation. This panel includes key phosphorylated and total proteins involved in NFκB signaling, such as IκBα, IKKα/β, and NFκB itself, along with upstream and downstream effectors like TNFR1 and c-Myc. Quantifying these analytes provides insights into cellular responses to stress, cytokines, and other stimuli in rodent models.

Application

MILLIPLEX® Qualified assays undergo rigorous assay development, verification, and Quality Control testing to achieve optimal performance. Simultaneously analyze 6 analytes in human cell lysate samples.Analytes Included: c-Myc, FADD (Ser194), IκBα (Ser32), IKKα/β (Ser177/Ser181), NFκB (Ser536), TNFR1Assay Characteristics: Refer to the assay protocol for details on cross-reactivity, buffer compatibility, lysate control, and recommended loading control (MAPmateControl Assays).

Features and Benefits

  • Measures six key signaling proteins involved in the NFκB pathway in cell or tissue lysates
  • Supports detailed analysis of phosphorylation events and pathway activation
  • Ideal for studies focused on inflammation, immune response, and cellular stress signaling
  • High sensitivity and reproducibility for reliable intracellular protein quantification
  • Offers a high-throughput alternative to Western blotting, reducing assay time and sample requirements while improving data consistency

"Toll-like Receptors (TLRs) are transmembrane proteins that are expressed on various immune cells. The extracellular N-terminal region of TLRs recognizes specific pathogen components. At least 13 different members of TLR family have been identified that detect different pathogen associated molecular patterns (PAMPs), including lipopolysaccharides, flagellin, bacterial CpG DNA, and viral RNA and DNA. Recognition of PAMPs by TLRs is considered as a key process for the induction of an inflammatory response."
